1. Anti-glycation effect of Cherry Flower Extract

Cherry Flower Extract has been proven as an anti-glycation agent that promotes collagen formation in fibroblasts that leads to anti-aging. To understand this, here is a short explanation of the mechanism of skin aging, leading to pigmentation, darkening and wrinkles.

Maillard reaction as source of aging

The Maillard reaction, named after L. C. Maillard, is also known as nonenzymatic browning. Maillard reaction is a chemical reaction between Amino Acids and reducing sugars by the impact of heat. Formation of AGEs has been found to contribute to the progression of age-related diseases and diabetes. The Maillard reaction often involves sugar binding proteins such as collagen and elastin. Glycation of collagen and elastin in the skin causes the accumulation of AGEs, leading to intracellular damage and programmed cell death.

Key role of AGEs and skin ageing

AGEs accumulation is fundamental in skin aging, as it leads to oxidative stress and inflammation.

AGEs are formed intracellularly and extracellularly. They can react with proteins, lipids and nucleic acids in almost all skin cells as well as on intracellular or extracellular proteins. Through alteration of the physicochemical properties of dermal proteins, decreased cell proliferation, increased apoptosis and senescence, induction of oxidative stress and proinflammatory mediators as well as other pathways, AGEs contribute to the overall picture of skin aging.

High levels of AGE can also damage the regeneration of the skin, induce injury through cross-link of collagen fibrils, which results with the hardening of the collagen matrix and finally leads to apoptosis or death of fibroblasts. As a result the dermis collagen is injured, and skin becomes wrinkled and dull.

Proteins with slow turnover rate, such as collagen as well as long-lived proteins, are primary targets of glycation reaction in the skin. Accumulation of AGEs due to glycation of collagen may induce programmed cell death in fibroblasts creating a negative effect on hyaluronic acid, collagen and elastin.

2. Bioactive ingredients of Cherry Flower Extract

A joint study by Kyoto Pharmaceutical University successfully identified caffeoyl glucose (1-caffeoyl-O-β-D-glucopyranoside) and quercetin glucoside (quercetin 3-O-β-D-glucopyranoside) as the bioactive ingredients of Cherry Flower Extract.

Inhibition of AGEs production – ANTI-AGING

Bioactive components of Cherry Flower Extract and raw Cherry Flower Extract were added to buffer a solution containing D-glucose and bovine serum albumin at 60°C and left to stand for 2 days. Raw Cherry Flower Extract (100μg/mL) significantly inhibited the production of AGEs.

Furthermore, the major bioactive ingredient, caffeoyl glucose remarkably inhibited the production of AGEs at concentration as low as 10μg/mL. The inhibitory effect of quercetin glucoside was double stronger than that of kaempferol glucoside.

Inhibition of AGEs production in fibroblasts – ANTI-AGING

Normal human fibroblasts were treated with glyoxal (glycated intermediate agent), incubated for 5 days and then skin glycation was analyzed by Western-blotting method. The result is that Caffeoyl glucose inhibited the production of AGEs at 1 and 10μg/ml, as quercetin glucose performed less efficient. Cherry Flower Extract at 10μg/ml significantly inhibited the production of AGEs promoted by glyoxal, compared with control group. Thus, it is evident, that caffeoyl glucose plays the major role in the inhibitory effect of CHERRY FLOWER EXTRACT on the production of AGEs.

Inhibition of fibroblasts apoptosis– ANTI-AGING

Accumulation of AGEs in skin activates the skin damage and the programmed cell death of fibroblasts. The effect of Cherry Flower Extract and its bioactive ingredients on carboxyl methyl lysine (CML) collagen induced fibroblasts apoptosis was tested. Cherry Flower Extract, caffeoyl glucose and quercetin glucose reduced caspase activity, resulting in inhibition of fibroblast apoptosis. The effect of caffeoyl glucose was extraordinary in the inhibition. This proves that the Cherry Flower Extract is potent and useful as an anti-ageing agent.

Promotion of collagen formation in fibroblasts, KEEPING THE SKIN YOUNG

The fibroblast is the principal active cell of the connective tissue that is responsible for making the extracellular matrix and collagen. Fibroblasts are involved in tissue metabolism and play an important repair role.

Fibroblasts were cultured in collagen containing medium and treated with glyoxal, a glycation inducer. Then it was observed, that when cultured like that, the Fibroblasts were regularly producing a collagen lattice. The growth of fibroblasts and collagen lattice formation were increased in the presence of Cherry Flower Extract of 100μg/ml and 1000μg/ml. Cherry Flower Extract prevents glycation of fibroblasts and to be efficient in maintaining extracellular collagen matrix.

Inhibition of melanin formation in B16 melanoma cells – NATURAL BRIGHTENING EFFECT

The skin brightening effect of Cherry Flower Extract was tested in B16 melanoma cells. Cherry Flower Extract was added to cell culture and incubated for 3 days. Then cells were crushed by extensive sonication termed “Hyper sonication” method, followed by absorbance measurement. The effect of the Cherry Flower Extract showed inhibition of melanin formation in a dose-dependent manner.

A double-blind, placebo-controlled study revealed that oral intake of collagen peptides significantly reduced wrinkles and increased the collagen in the skin.

More than 100 women aged between 45 and 65 years were randomized to receive 2.5 g of collagen peptides or placebo, once daily for 4 weeks.

The study showed that the oral intake of collagen peptides (2.5 g/day) significantly reduced wrinkles after 4 weeks and lead to a remarkably higher content of procollagen in the skin.

Cellulite is not a medical issue. Cellulite is a purely cosmetic condition that many women suffer from, as it affects 80-90% of women worldwide.

Supplementation with collagen peptides also reduced the skin waviness, with first results visible after 3 months, with even more explicit results after 6 months. Collagen peptides treatment with daily dose of 2.5 g led to a statistically significant decrease in the degree of cellulite and reduced skin waviness in women with a BMI < 25. Moreover, dermal density was remarkably improved (P < 0.05) compared to placebo.

The results of the study demonstrated that a long-term therapy with orally taken collagen peptides leads to an improvement of cellulite and has a positive impact on skin health.

Additionally, compared to the control group, the nail growth speed was increasing. Collagen peptides led to a statistically significant improvement in nail growth after 3 months of treatment. At the same time, there was a remarkable improvement in nail peeling and a clearly decreased nail edge irregularity.

80 % of the participants agreed that the use of collagen peptides improved their nails’ appearance and were completely satisfied with the performance of the treatment. 75 % of the women perceived their nails as longer, 71 % said their nails grew faster and became longer.

The results of the study showed that a long-term therapy with orally taken collagen peptides leads to a faster growth, decreases nail roughness and peeling and has a significant beneficial effect on the overall health and outlook of the nails.
